Have denuding handles nearly replaced the old school pulled pipettes?

With stupendous progress of ART in the last decade, the corresponding success rate has steadily climbed. Many improvisations have added impetus. Since I started training as an Embryologist, the onus I comprehend in the flow of IVF process beginning with ovum pick and ending with embryo transfer is the transportation of oocytes, zygotes, and Embryos across micro drops, Petri dishes, wells, and Nunc plates. This transportation involves incorporation of embryos into the fresh culture from the previously spent culture dish, lodging freshly denuded oocytes into micro drops of ICSI plate and also is instrumental for conveyance during cryopreservation procedures. Embryologists can choose from traditional pulled Pasteur Pipettes but undoubtedly, denuding handles or devices have become a norm.

Cotton plugged Pasteur pipettes are basically the original vectors for handling embryos and oocytes. Glass made, they exorbitantly are cheap which serves as its biggest advantage. But they have to be made by ”pulling” them in flames. The process of pulling a standard Pasteur pipette and converting it into an IVF or an ICSI pipette essentially needs to be skillful in lieu with diameter size and the fate of the surface. A rough or daggered edge has the potential to damage the embryos and the oocytes. A lot of precision is supposed to be invested in pulling these pipettes demanding practice and control. Moreover, there are sterility issues too!

On the other hand, a denuding device is kind of an adjustable handle; the basic primary anchoring structure on which ‘denuding pipettes or flexipets ’ are lodged at its tip. These instruments can predetermine a specific pool of media it can aspirate which is wholly 0.25 µl to 3.0 µl. The cost factor may put glass pipettes in feasibility zone; however, denuding devices entail the liberty of not requiring pulling and making them. The flexipets which are lodged at the tip and are disposable come in different sizes catering to various procedures and stages of the embryo.

‘Control’ over embryos and oocytes is effective and standardized in denuding handles but not in glass pipettes. Volume predetermination in lieu with the effective bending of flexipets makes them look better over glass pipettes which have a flipside of ‘no bend-break trend’ coupled with a tremendous emphasis on controlling media comprising of embryos. If embryos or oocytes lose their way in a denuding handle, a vigorous ‘’pipetting’’ can help you recover them back. But in case of a pulled glass pipette, there is a monumental risk of losing them forever.

EZ-Grip from RI (Research Instruments) consumables promoted by Shivani Scientific Industries Pvt Ltd has all the hallmarks of a good denuding handle which is laced with features such as precise control system, pipette size indicator and is readily autoclaved. This device also comes with EZ-Tip (disposable tips) which is sold in two options of different packaging systems. They come in different sizes from 125-600um and is flexible, shatterproof medical grade plastic, MEA, LAL tested and effectively sterilized.

It is important to consider an effective denuding device for procedures such as denudation and cryopreservation considering it is tedious and may require multiple rounds of transportation.
Very few clinics now must be using pulled pipettes and it is a matter of time before they may get completely obsolete with respect to IVF!
