Biopolymer-based Nanosystems for the Treatment of Kidney Diseases

AKI occurs in approximately 10-15% of patients following hospital admission and has a high mortality rate. CKD, on the other hand, is defined by a decrease in glomerular filtration rate and affects millions of patients worldwide. There is an urgent need for new treatments for kidney disease.

Nanomedicine, which involves the use of nanoparticles, is a promising field for treating and diagnosing diseases. Nanoparticles have unique properties that make them suitable for drug delivery systems. They can enhance drug stability, solubility, and transport across cell membranes. Moreover, nanoparticles can concentrate drugs at the site of disease while minimizing their adverse effects. Various nanoparticles have been used as carriers for kidney-Targeted Drug Delivery.

Biopolymers, which are polymers produced by living organisms, show great potential when combined with nanotechnology for targeted drug delivery. They have better biocompatibility, bioavailability, and bioreactivity compared to synthetic polymers. Biopolymer-based nanosystems have been particularly promising for the treatment of kidney diseases.

One strategy for targeted drug delivery to the kidney is through the use of megalin, a receptor protein expressed in renal proximal tubular epithelial cells. Megalin can bind to various ligands, including drugs, and mediate their endocytosis into the cells. Researchers have successfully coupled megalin-binding substances like lysozyme, chitosan, and polymyxin B to drugs for kidney-specific delivery. These conjugates have shown promising results in targeting the renal tubules and treating kidney diseases.

In conclusion, biopolymer-based nanosystems hold great potential for the treatment of kidney diseases. Targeted drug delivery strategies, particularly using megalin as a receptor, have shown success in concentrating drugs in the kidney and treating renal diseases effectively.
